Any image that has or deserves the [edit] tag should probably have largely identical tags to its source image.

_{EDIT: I will add the caveat that I, personally at least, generally don’t aim for quite as much tagging detail when it comes to meme edits.}

When I’m tagging, my two main focusses are increasing the image’s searchability and filterability.

My basic process looks something like this:

Character gender?
[male] [female]

How many characters?
[solo] [duo] [trio] [group]

Tag for whether the duos/trios contain multiple genders.
[duo male] [duo female] [duo male and female] [trio male] [trio female] [trio male and female]

Is there any shipping or romance happening?
[shipping] [straight] [lesbian] [gay] [there’s also probably a dedicated tag for whatever ship is displayed]

What are the body types of the characters?
[pony] [anthro] [semi-anthro]

If pony, what species?
[earth pony] [unicorn] [pegasus] [alicorn]

Are they fully grown, or smaller?
[stallion] [mare] [foal] [colt] [filly]

What kind of background?
[flat background] [transparent background] [abstract background] [gradient background]

If not just a background, what’s the setting?
[indoors] [outdoors]

Those are the main things I try to get with most every image I tag on. There’s plenty of other stuff to pay attention to, though:

Where are each of the characters looking?
[Looking at you] [looking at something] [looking at someone] [looking at each other] [looking away] [looking up]

If [looking at each other] also check for [eye ].

Consider describing the eyes as well:
[catchlights] [no catchlights] [eyes closed] [wide eyes]

I only ever tag [eyes open] if there’s multiple characters and only some of them have [eyes closed].

What are the expressions?
[happy] [sad] [smiling] [frowning] [staring] [open mouth] [closed mouth]

Is someone [talking]? If there’s readable text, also use [dialogue].

What are each of the character’s stances?
[flying] [jumping] [running] [walking] [standing] [sitting] [lying down] [prone]

How are they using their limbs?
[raised hoof] [raised leg] [pointing] [waving]

Tag extra appendages on pony species:
[horn] [wings]

How are pegasus wings positioned?
[folded wings] [partially spread wings] [spread wings]

Is a pony using magic?
[magic] [telekinesis]

Is anypony [holding] anything? [Eating] [food]? [Drinking] a [drink]?

… And many, many more. Any proper tagging assistance is always appreciated, but honestly, just getting the most important tags will still make everypony happy.
